چکیده

A system for plant regeneration from individual cells or explants is essential for the application of genetic engineering. In Cucurbitaceae, in vitro regeneration has been reported across a wide spectrum of crops, including summer squash (Cucurbita pepo L.) (7), winter squash (Cucurbita maxima Duch.) (9), bottle gourd (Lagenaria siceraria Standl.) (6) and, recently, figleaf gourd (Cuburbita ficifolia Bouché.) (8). Genetic transformation was applied in this family for different purposes, for example to improve several cucurbits with potential to function as rootstocks (4, 6, 11). Regeneration ability is highly influenced by genotype, and then it is necessary to develop and adjust the appropriate regeneration protocols for each cultivar. In this work we assay different in vitro conditions useful for regeneration in eight Cucurbita cultivars which presented interesting characteristics for breeding. They include: three C. pepo, two C. ficifolia one C. maxima and two C. moschata species. To the best of our knowledge this is the first study reporting in vitro regeneration for the latter species.

High Throughput Sequencing of Small RNAs in the Two Cucurbita Germplasm with Different Sodium Accumulation Patterns Identifies Novel MicroRNAs Involved in Salt Stress Response

MicroRNAs (miRNAs), a class of small non-coding RNAs, recognize their mRNA targets based on perfect sequence complementarity. MiRNAs lead to broader changes in gene expression after plants are exposed to stress. High-throughput sequencing is an effective method to identify and profile small RNA populations in non-model plants under salt stresses, significantly improving our knowledge regarding ...

Assessment of Genetic Variation in Twelve Accessions of Tropical Pumpkin (Cucurbita maxima) of South Eastern Nigeria

Submitted: Mar 17, 2013; Accepted: May 1, 2013; Published: Aug 5, 2013 Abstract: Twelve accessions of tropical pumpkin of South Eastern Nigeria were evaluated in 2010 and 2011 to determine the extent of variability that exit in fruit, seed and leaf yield of the crop and to constitute a germplasm collections. Results of the study show that there exists a sufficient genetic variability among the ...

Response of squash (Cucurbita spp.) as rootstock for melon (Cucumis melo L.)

The effects of different Cucurbita spp. rootstocks on melon cultivars belonging to different horticultural groups were studied by comparing grafted plants with non-grafted ones under greenhouse and open ®eld growing conditions. Winter melons (Cucumis melo L.) of the Inodorus group, cvs. `Thraki', `Peplo' and `Lefko Amynteou', and a summer melon of the Cantalupensis group, cv. `Kokkini Banana', ...

In vitro Plant Regeneration of Helianthus Annuus (Hyb. Azargol) From Alginate-Encapsulated Shoot Tips for Short Term Storage, Germplasm Exchange and Distribution

The present study demonstrates the potential of nutrient-alginate encapsulation of shoot tips of sunflower, Helianthus annuus (hyb. Azargol) for synthetic seed technology, which could be useful in germplasm distribution and exchange. Shoot tips from in vitro shoot cultures derived from mature seed explants were encapsulated in 3% sodium alginate and 100 mM CaCl2. 2H...

Translocation of structural P proteins in the phloem.

Phloem-specific proteins (P proteins) are particularly useful markers to investigate long-distance trafficking of macromolecules in plants. In this study, genus-specific molecular probes were used in combination with intergeneric grafts to reveal the presence of a pool of translocatable P protein subunits. Immunoblot analyses demonstrated that Cucurbita spp P proteins PP1 and PP2 are translocat...
